Training for 2 day bike ride

We are considering doing the 2 day bike ride for MS. Problem is,the ride is in October and we would all need to major training since we have not been on a bike in quite some time. We also have not done any type of bike event before. Is it possible to get ready for something like this before October with training on the road combined with spin class?

Absolutely.  One of the nice things about a multi-day event is that although each day has what looks like a lot of miles, when you space it out over the day, it really isn't too far.  Though I don't know your fitness, I believe your most critical need is to build your tolerance of time in the saddle.  If I were to set up a training schedule, it would start with easy spinning for increasing periods of time.  Start at 30 minutes easy, then work your way up to 1 hour in the first month, riding 3-4 times a week (on the spin bike or, better, outdoors) and concentrating on smoothness and quick cadence in pedal stroke.  Once you are comfortable with one hour, you can begin working harder at spin sessions and throw in a long ride (1-2 hours) on the weekend each week.  By September, you will be surprised at how fit you are getting and you may be able to increase your weekend sessions to 2-4 hours.  Once you have 4 hours in the saddle comfortable you should be able to do the all-day, on and off the bike of a multi-day.
What I would warn against is trying to do it all at once.  You don't want to get on the bike and work so hard it is unpleasant, because that will just train you to dislike your bike sessions.  Take your time, get on the bike frequently and realize that you have plenty of time to build your fitness.
And ENJOY!  You will see some wonderful things happen to your fitness if you make it a part of your lifestyle in the coming months.
